چکیده

Camels represent part of the Arab heritage. The interest in developing assisted reproductive technologies and cryobanking for the conservation of animal genetic resources has recently increased. However, semen collections in camelids present many problems as sitting position during copulation, slow ejaculation and difficult animal handling. In these cases epididymal sperm from slaughtered or recently died animals will increase the opportunities to create semen storages. The present work was designed to assess motility of camel epididymal sperm extended in Ovixcell(r) and in Tris-fructose-egg yolk semen extender. Spermatozoa were extracted from 16 epididymides using the retrograde flushing technique, washing sperm cells in a retrograde direction from the ductus deferens through the cauda epididymidis with a syringe loaded with warmed (37°C) extender. Total motility was evaluated after 15 minute of incubation in a water bath at 37°C under phase-contrast microscopy using a pre-warmed (37°C) Makler Chamber. Total motility was similar in Ovixcell(r) or Tris-fructose-egg yolk semen extender (52.8 ± 0.7% vs 41.22 ± 33.56%, respectively). Further studies aiming to the test the fertilizing capacity should be carried out in order to confirm the optimal testicles storage condition for the creation of semen cryo storages in camels.

Developmental competence of Dromedary camel oocytes fertilized in vitro by frozen-thawed ejaculated and epididymal spermatozoa

The present study aimed to compare the in vitro fertilizing capacity of frozen-thawed ejaculated and epididymal spermatozoa in order to standardize the semen preparation protocol for camel in vitro fertilization (IVF). Semen samples were collected from 7 Dromedary camels by means of artificial vagina (AV). Ten cauda epididymes were obtained from slaughtered adult camels, isolated, incised and r...

Assessment of epididymal sperm obtained from dromedary camel

Testicles were isolated from dromedary camels in a local slaughterhouse at breeding and non-breeding seasons. Sperms were recovered from different parts of the epididymis (caput, corpus and cauda) and stained separately on slide glasses by eosin nigrosin staining method and dried by a hair dryer and carried to the laboratory. In the lab, slides were observed for evaluation of the proportion of ...

Effect of testicle postmortem storage on goat frozen-thawed epididymal sperm quality as a tool to improve genebanking in local breeds.

The interest to develop assisted reproductive technologies and cryobanking for farm animal genetic resource conservation has recently increased. However, cryopreservation for ex-situ management of genetic diversity sometimes is not routinely feasible, owing to the lack of facilities (AI centres, laboratories) and expertise near the local breed farming area. In these cases, epididymal sperm obta...

P-34: Effect of Glucose on Buffalo Epididymal Sperm Motion Characteristics, Viability and Cytoplsmic droplets During 24 Hours Incubation

Background: The recovery of sperm from the cauda epididymis may be the last chance to conserve, genetic Materials of valuable farm animals. Sperm utilize glucose as source of energy through glycolysis and oxidative phosphorylation to provide motility and movement. While endogenous glucose of the culture media has little potential to preserve sperm motility and viability for long period of time ...
